It acts as a potent inhibitor of dihydrofolate reductase (DHFR), an enzyme critical for folate metabolism and DNA synthesis. By blocking this enzyme, methotrexate disrupts DNA synthesis and cellular replication, making it effective against rapidly dividing cancer cells.
